PNPT1/SUV3 knockdown induces dsRNA accumulation in surrounding fibroblasts but not in dispersed human islet cells. Dispersed human islets were transfected with a siRNA control (siQ) or with siRNAs targeting PNPT1 (#1), SUV3 or both (P/S), and then maintained in culture for 6 days after transfection. dsRNA accumulation (red), insulin content for beta cell staining (green), and vimentin for fibroblast staining (purple) were analyzed by immunocytochemistry.
